Fig. 8. Anti-CD3/anti-CD28-expanded cells express less Siglec-5 than nonexpanded T cells. Unstimulated monocyte-depleted PBMCs were mock-transfected with no DNA or transfected with 1, 2, or 3 mg of pSig5 by using the Amaxa Nucleofector device. Transfected cells were stimulated with anti-CD3/anti-CD28-bearing beads for 3 days. Cells were then analyzed for expansion based on side and forward scatter and the expression of Siglec-5. Side and forward scatter dot-plot profiles of anti-CD3/anti-CD28 bead-stimulated mock-transfected cells (A) and 3-mg pSig5-transfected cells (B) are shown. (C) Mean fluorescence intensity of Siglec-5 expression of the unexpanded and expanded cells after stimulation for each sample is shown.
