Unstimulated monocyte-depleted PBMCs were mock-transfected with no DNA or transfected with 1, 2, or 3 mg of pSig5 by using the Amaxa Nucleofector device. Transfected cells were stimulated with anti-CD3/anti-CD28-bearing beads for 3 days. Cells were then analyzed for expansion based on side and forward scatter and the expression of Siglec-5.
